read my game informer. I also tried to summarize it here, but failed, so here is a summary of that summary.
10 years after first game
you are a renegade BigDaddy
you can use plasmids
you can use plasmids at the same time as your guns.
You get a cool drill
A little sister from the first game has come back to rapture as a big sister
she happens to be your main enemy
and after you kill another big daddy, you can trick their little sister into thinking that you are theirs, and you can protect them while they harvest adam for yyou.

Of Course you didn't explore all of Rapture, so there will probably be plenty of other whack jobs you get to meet who will contribute to or hinder your progress.
